SPINeasy® 96-Well DNA Pro Kit for Soil
The SPINeasy 96-Well DNA Pro Kit for Soil has been developed to accommodate a wide range of soil types. Performance testing demonstrates its efficacy in extracting nucleic acids from soils with varying biomass levels, ranging from high to low. The kit provides optimal yield and purity for various types of soil samples. It displays an exceptional ability to extract gDNA from different types of soil. The SPINeasy 96-Well DNA Pro Kit for Soil consistently delivers higher amount and purity of DNA as compared to the commercially available competitors A and B kits (Figure 1, upper). The extracted DNA samples were analyzed using Agilent Tapestation 4150 to examine their integrity and yield. The DNA integrity (DIN) values were comparable for samples extracted from the three kits; nonetheless, the nucleic acid yield was higher when extracted with the SPINeasy 96-Well DNA Pro Kit for Soil, as indicated by the more intense bands on the virtual gels. The extraction performance is significantly better than that of competitors. (Figure 1, lower). The SPINeasy 96-Well DNA Pro Kit for Soil stands out for its ability to consistently extract high-quality genomic DNA (gDNA) from diverse soil samples, surpassing the performance of competitors’ products.
Figure 1. DNA quality. (Upper). Agarose gel electrophoresis of gDNA extracted using SPINeasy 96-Well DNA Pro Kit for Soil and competitor kits. The respective DNA yield and purity ratios (A260/280 and A260/230) were assessed using spectrophotometer. Each dot of the plot represents a single extraction. The horizontal bars indicate the median value. (Lower). Virtual gels obtained using the Agilent 4150 TapeStation analyses were shown along with the DNA integrity (DIN) values. Soil samples with various biomass content (250 mg each) were processed using SPINeasy 96-Well DNA Pro Kit for Soil, competitor A, and competitor B kits.
Genomic DNA extracted from soil, heavily contaminated with humic acid, using SPINeasy 96-Well DNA Pro Kit for Soil could be readily used in quantitative PCR (qPCR) with no inhibition observed (Figure 2). This suggests the effectiveness of the kit in removing PCR inhibitors. qPCR results of gDNA extracted using SPINeasy 96-Well DNA Pro Kit for Soil showed lower CT values as compared to the competitor kits (Figure 2).
Figure 2. Amplifiability. qPCR amplification data of gDNA extracted from three soil samples using SPINeasy 96-Well DNA Pro Kit for Soil and competitors’ kits. DNA template amount: 50 ng. Primers: 16S rRNA (197 bp). The absence of inhibitor was assessed using inhibitor-sensitive quantitative PCR and undiluted samples. The bacterial 16S rRNA gene was amplified using gDNA extracted from high biomass soil. This extraction was performed using the SPINeasy 96-Well DNA Pro Kit for Soil and kits from other competitors. As the gDNA extracted with SPINeasy 96-Well DNA Pro Kit for Soil could successfully be used in downstream applications (such as qPCR), this demonstrated the effectiveness of the kit in removing the inhibitors.
