Difficulty in RNA extraction from Mycobacterium

Like any RNA, the RNA in Mycobacterium is typically unstable and prone to degradation. Furthermore, the high lipid content in cell wall increases the difficulty of RNA extraction from Mycobacterium.

Overview

Sample Name:

Mycobacterium smegmatis

Sample Type:

Mycobacterium culture

Challenge :

RNA extraction from Mycobacterium smegmatis was unsuccessful.

Featured Materials:

Methods

Sample was prepared in order of protocol in accordance to product guidelines found here.

Based on MP company’s technical advice, we optimized experiment conditions.

Outcome

Trial results data

Electrophoresis chart

The extracted RNA was evaluated in a Nanodrop ND-1000 spectrophotometer.
The acceptable ranges of absorbance ratios at 260/280 and 260/230 were 1.8-2.0 and 1.8-2.2 respectively

Conclusion

Key takeaways:

Based on MP company’s technical advice, we optimized the experiment conditions, and both the RNA yield and purity were improved. The RNA extracted from the Mycobacterium smegmatis can meet downstream experiment requirements. The researcher was satisfied with the results and has already made SPINeasyTM RNA Kit for Bacteria as his choice for future studies.
