Analysis of prokaryotes such as bacteria from environmental water needs a different approach
The microflora in environmental water consists of a high density and diversity of bacterial species. The composition of the microflora from the environmental water such as waste water, pipe water, river water and marine water is dependent on various conditions such as temperature, salinity, organic or inorganic nutrients and human activities. However, majority of these species cannot be cultured in vitro, so an alternative approach is needed.
The alternative approach would be to conduct metagenomic analysis of the environmental water samples. For analysis of the DNA sequences, genomic DNA would need to be extracted from the environmental water through a sample preparation process which for water samples would typically involve the first step of membrane filtration to collect the microbials, followed by cell lysis to release the nuclei acid contents and then to end with nuclei acid isolation and purification. The DNA content is then be sequenced and analysed for its metabolic profile.
